BNP和NT-proBNP


脑钠肽(BNP ) 是一种具有利钠、舒张血管及抑制肾素功能的多肽类激素。脑钠肽属于一个包括ANPCNP及尿舒张肽的结构类似多肽家族。这些多肽的特点是具有一个17个氨基酸通过一对二硫键组成的环状结构。环状结构在不同的利尿钠肽之间表现出高度一致性(全族多肽环状结构的17个氨基酸中有11个是相同的)。BNP分子由32个氨基酸残基组成,包括一个由Cys10Cys26通过二硫键所组成的环状结构。

 

    BNP是脑钠肽原前体(preproBNP)的蛋白酶裂解产物。preproBNP含有134个氨基酸残基,在心肌细胞内合成。preproBNP脱去信号肽(1-26位氨基酸残基)后成为脑钠肽前体(proBNP)分子(27-134位氨基酸残基)。 proBNP108个氨基酸残基)在转化酶依赖反应中形成两个多肽,BNP77-108位氨基酸残基)和proBNPN端部分(NT-proBNP1-76位氨基酸残基)。BNP(具有生物 活性)和NT-proBNP(生理活性不确定)以及没有分解的 proBNP都被分泌进入外周血。这种proBNP不完全分解的原因直到最近才清楚。
